The destinations" respective tourist boards met at the INDABA travel industry event this week to initiate the deal.
All of the organisations agreed on the importance of city tourism, stressing that cities are the most important and influential brands for the majority of countries.
Mariette du Toit-Helmbold, chief executive of Cape Town Tourism, said: "Cape Town, Johannesburg and Durban tell three very different and unique but complementary stories.
"I strongly believe that we add value to each other"s leisure and business tourism offering, and I think that this will also go a long way towards providing the prospective international traveller with a seamless impression of destination South Africa."
Johannesburg Tourism chief executive Lindiwe Kwele said each of the destinations can be defined and promoted on the strength of its individual traits.
She described Johannesburg as a city characterised by "urban chic" and a "sophisticated energy"; Cape Town as a place of "inspiration, hope [and] unprecedented natural beauty"; and Durban as a "leading port city" full of warmth and tradition.
